(AP) ? Andy Reid's worst coaching season with the Philadelphia Eagles ended Monday after 14 years when he was fired by owner Jeffrey Lurie, who said it was time "to move in a new direction."

The dismissal came one day after Reid and the Eagles were humiliated 42-7 by the New York Giants and ended their season at 4-12.

"Andy leaves us with a winning tradition that we can build upon. And we are very excited about the future," Lurie said in a statement released by the team.

The team pushed back a scheduled noon EST news conference to 1 p.m.

Reid took over a 3-13 team in 1999, drafted Donovan McNabb with the No. 2 overall pick and quickly turned the franchise into a title contender.

He is the winningest coach in club history and led them to a run of four straight NFC championship games, a streak that ended with a Super Bowl trip after the 2004 season ? and a loss, 24-21, to the New England Patriots.

Still, Reid cemented Philadelphia as a football town ? though the Eagles have never won the NFL title ? and led the team to an unmatched level of success. But the team hasn't won a playoff game since 2008 and after last season's 8-8 finish, Lurie said he was looking for improvement this year.

Instead, it was even worse.

Reid sounded like a man who knew he was going to be out of work when he addressed the media after the Giants game.

"I go in eyes wide open," Reid said, referring to his upcoming meeting Monday with Lurie. "Either way, I understand. Whatever he chooses will be the right thing. He always does things for the best interests of the Eagles."

Lurie said the search for Reid's successor begins immediately.

Reid is due to make $6 million in 2013 in the final year of his contract. He is the franchise leader in wins (140) and winning percentage (.578) and led the Eagles to six division titles and five NFC championship games.

It already had been a difficult year for Reid. He endured a devastating loss weeks before the season opener when his oldest son, Garrett, died at training camp after a long battle with drug addiction.

In October, Reid fired close friend and longtime assistant Juan Castillo, who was in his second season as defensive coordinator after coaching the offensive line for 13 years. He later fired defensive-line coach Jim Washburn.

After beating the defending Super Bowl champion Giants on Sept. 30, the Eagles lost eight straight games ? their worst losing streak in 42 years.

PhiladelpiaEagles.com posted video of Lurie and Reid addressing team employees, who gave Reid a big ovation. Lurie handed him a game ball.

"I have a hard time standing before people without a few boos involved. But I'm taking it, I'm taking it all in," Reid said. "These have been the greatest 14 years of my life."

He added: "Sometimes change is good. ... I know the next guy that comes in will be phenomenal. The ultimate goal is a Super Bowl. Everybody in this room, I wish you a big ring on the finger in the near future.

"Hail to the Eagles, baby."

'Downton Abbey' Spoilers: Creator Julian Fellowes On Dan Stevens' Exit In Christmas Special

SPOILER ALERT -- do not read on if you have yet to see the 2012 "Downton Abbey" Christmas special, which aired in the UK on December 25.

The halls of "Downton Abbey" will be a little emptier next season. Earlier this week, viewers were shocked when it was confirmed that Dan Stevens (Matthew Crawley) would not be returning for Season 4 of the period drama, after his character definitively exited the series in this year's Christmas Special.

Now, "Downton" creator Julian Fellowes has weighed in on the actor's abrupt exit, telling British newspaper the Daily Telegraph, "We have always tried to persuade him to stay. He had done fantastically well for the series and his creation of Matthew was terrific. But this is life. Dan felt that this was right thing for him, and the right moment to move on to different areas."

Stevens' exit proved particularly controversial, considering that Matthew was killed in the Christmas episode, rather than the door being left open for a future return. According to the Telegraph, "Downton's" British broadcaster, ITV, has since been inundated with complaints that the storyline was "a 'travesty', a 'tragedy' and an 'outrage', with some saying the ending had 'ruined their Christmas'."

But Fellowes maintained that killing Matthew was really the only course that the show could take once Stevens decided to depart. ?If he had been prepared to come back for maybe two or three episodes in a series, that would be different. Then we could have had a foreign posting or invented a career that would have made it possible for him to be away," the creator said. "Otherwise we would have had to make this tremendously successful love affair between Mary and Matthew unhappy, which I didn?t feel would be believable ... So we didn?t really have any option. By him dying, their love can remain in tact."

While Season 3 has yet to air on PBS in the US, rumors of Stevens' exit began circulating as early as March, when it was revealed that Stevens, Jessica Brown Findlay (Lady Sybil) and Siobhan Finneran (O?Brien) hadn't renewed their contracts past Season 3. The speculation began anew after the actor relocated to New York to star in "The Heiress" on Broadway. "New York for me is going to be one big adventure," Stevens said at the time. "I will be taking the whole family out and we will be there for six months. I don?t know if I will be returning to 'Downton.'"

After the episode aired, Stevens explained his decision to exit the show. ?We were always optioned for three years,? he told the Telegraph. ?And when that came up it was a very difficult decision. But it felt like a good time to take stock, to take a moment. From a personal point of view, I wanted a chance to do other things."

ITV then released a statement on Stevens' departure in an attempt to console the show's heartbroken audience: "After three successful series and two Christmas editions of 'Downton Abbey,' Dan Stevens decided not to renew his contract beyond the initial three years he had been contracted ... Over the last three years, audiences across the world have been captivated by the ups and downs of Mary and Matthew?s relationship, culminating in their wedding. Fans have enjoyed what has become a solid and loving marriage. It is for this reason that the Producers decided Matthew and Mary could not simply be estranged or parted, resulting in his untimely and tragic death at the end of the Christmas episode."

Praying Hitler in ex-Warsaw ghetto sparks emotion

WARSAW, Poland (AP) ? A statue of Adolf Hitler praying on his knees is on display in the former Warsaw Ghetto, the place where so many Jews were killed or sent to their deaths by Hitler's regime, and it is provoking mixed reactions.

The work, "HIM" by Italian artist Maurizio Cattelan, has drawn many visitors since it was installed last month. It is visible only from a distance, and the artist doesn't make explicit what Hitler is praying for, but the broader point, organizers say, is to make people reflect on the nature of evil.

In any case, some are angered by the statue's presence in such a sensitive site.

One Jewish advocacy group, the Simon Wiesenthal Center, this week called the statue's placement "a senseless provocation which insults the memory of the Nazis' Jewish victims."

"As far as the Jews were concerned, Hitler's only 'prayer' was that they be wiped off the face of the earth," the group's Israel director, Efraim Zuroff, said in a statement.

However, many others are praising the artwork, saying it has a strong emotional impact. And organizers defend putting it on display in the former ghetto.

Fabio Cavallucci, director of the Center for Contemporary Art, which oversaw the installation, said, "There is no intention from the side of the artist or the center to insult Jewish memory."

"It's an artwork that tries to speak about the situation of hidden evil everywhere," he said.

The Warsaw ghetto was an area of the city which the Nazis sealed off after they invaded Poland. They forced Jews to live in cramped, inhuman conditions there as they awaited deportation to death camps. Many died from hunger or disease or were shot by the Germans before they could be transported to the camps.

The Hitler installation is just one object in a retrospective of Cattelan's work titled "Amen," a show that explores life, death, good and evil. The other works are on display at the center itself, which is housed in the Ujazdowski Castle.

The Hitler representation is visible from a hole in a wooden gate across town on Prozna Street. Viewers only see the back of the small figure praying in a courtyard. Because of its small size, it appears to be a harmless schoolboy.

"Every criminal was once a tender, innocent and defenseless child," the center said in a commentary on the work.

Poland's chief rabbi, Michael Schudrich, said he was consulted on the installation's placement ahead of time and did not oppose it because he saw value in the artist's attempt to try to raise moral questions by provoking viewers.

He said he was reassured by curators who told him there was no intention of rehabilitating Hitler but rather of showing that evil can present itself in the guise of a "sweet praying child."

"I felt there could be educational value to it," said Schudrich, who also wrote an introduction to the exhibition's catalogue in which he says art can "force us to face the evil of the world."

On Friday, a stream of people walked by to view the work, and many praised it.

"It had a big emotional impact on me. It's provocative, but it's not offensive," said Zofia Jablonska, a 30-year-old lawyer. "Having him pray in the place where he would kill people ? this was the best place to put it."

Cattelan caused controversy in Warsaw in 2000 when another gallery showed his work "La Nona Ora" ? or "The Ninth Hour" ? which depicts the late Pope John Paul II being crushed by a meteorite. That offended many in Poland, which is both deeply Catholic and was John Paul's homeland.

Iraqi Sunnis demonstrate in Anbar, west of Baghdad

FALLUJAH, Iraq (AP) ? Witnesses say tens of thousands of Iraqi Sunni Muslims have massed along a major western highway in the latest of a week of large demonstrations against the Shiite-led government.

The protesters gathered Friday near the city of Fallujah, which is in the vast Sunni-dominated province of Anbar west of Baghdad.

Protesters held aloft placards declaring the day a "Friday of honor." Some carried old Iraqi flags used during the era of ousted dictator Saddam Hussein. Others raised current ones.

The demonstrations follow the arrest last week of 10 bodyguards assigned to Finance Minister Rafia al-Issawi, who comes from Anbar and is one of the central government's most senior Sunni officials.

The province has been the scene of several large demonstrations and road blockages since last Saturday.

Back Pain Solutions(NaturalHealth365) To reduce pain within your body ? you?ll have to fix bad posture as soon as possible. Are you too tired to sit up straight? Well, slouching is actually harder work than sitting or standing with a naturally aligned spine.

When we slouch, we enlist muscles to perform functions they are not designed to do. Superficial muscles in our back and neck area become strained, tense and over-stretched. These muscles are designed to rotate the head and flex the neck, but not for long sustained contractions involved with alignment.

The hidden dangers of sitting (too much) at a computer

Deeper slow twitch postural muscles should do the work of holding our head up and our body graceful, powerful and pain-free. Slouching is an epidemic in the Western World, and I feel it is primarily a result of too much sitting poorly in chairs and hunching over laptops.

Our young people have the worst posture of any generation and x-rays of some 20 year olds look like the arthritic spines of older people. We need to take serious measures to teach good posture, provide ergonomic chairs in our schools and stop slouching.

Are government health officials really interested in saving money on healthcare?

Chronic pain costs billions every year taxing our health care system and the economy and much of the pain is a result of slouching when sitting in chairs and cars.

The chair has an unnatural right angle shape that does not conform to our naturally curving spine shape. Many people slouch in a chair and roll back and sit on the lower tailbone and sacral area instead of aligning their hips and ribs so they can balance on the sit bones at the base of the pelvis.

When we let the pelvis tilt backward, the breastbone sinks, the head goes forward and the neck and back muscles get drafted into dysfunctional habits. The diaphragm gets compressed and the movements of breathing are forced into the upper shoulder area that in many cases will enlist a low level flight or freeze response in the nervous system.

Poor posture contributes to chronic disease

The chronic adrenal fatigue created by slouching can eventually lead to chronic fatigue, fibromyalgia, rheumatoid arthritis, and many other autoimmune diseases.

Another bane of slouch sitting in chairs happens because we lose the tone of our core postural muscles, since the back of the chair does the work of keeping the spine upright. We get up out of our chairs but the chair habits stay in our body leading to chronic slouching.

Slouching is a result of dysfunctional breathing habits created by poor posture and also poor posture creates dysfunctional breathing habits. They are one in the same and the connection between the two is a huge blind spot in the medical, yoga and fitness world.

Stop and take a moment to breathe

One can avoid slouching by learning powerful breathing practices that engage the muscles that support the spine and diaphragm.

Exercising good posture involves waking up the core breathing muscles. Tight abs and shoulders drawn back are inefficient ways to stop slouching that can actually drain energy and restrict movement. Try holding your stomach in or pulling back on your shoulders while walking ? you can see and feel why this is not a good way to get aligned with better posture.

Many yoga poses and exercises engage our muscles to make us slouch!

Some athletes exercise in body positions that are a muscled up slouch position. Yogis do forward bends that shorten the front and overstretch the back, compressing the diaphragm and making the ligaments in the hips and spine too lose. Many people engage daily in ab crunching moves that enlist the muscles of the trunk to be short and tight which can cause the organs to protrude and make a ?pot belly.?

Cyclists in particular are known for hunching over their handlebars, and paddlers ? seated as well as stand up ? are often in very poor postural alignment when they exercise. The muscles that people engage to shorten the abdominals and round the upper back in these positions mimics slouching. Working the body hard in these positions engages the deep exhalation muscles and locks them in, so it is like they are literally stuck in the exhale.

Is the fitness industry really helping us to get fit?

Many fitness or yoga exercises that direct us to keep the belly tight and flat unnaturally stifle the breathing process, while shortening the front of the body and pulling the head and shoulders forward into a slouch.

Try taking a deep coughing action and then hold the action of these muscles to get an experiential feeling for what happens when our abdominals are trained to shorten. Exercising with our natural spine curves reversed or compressed restricts breathing, and forced tight abs can inhibit downward contractions of our diaphragm.

When exercising, doing things like keeping your back flat, tailbone tucked, belly tight, knees straight, or pushing belly out on inhale will actually inhibit your ability to breathe and can lead to more slouching.

How can we stop slouching?

Practice self-massage daily on the head, neck, feet, and arms to release unnecessary tension. Don?t wait for someone else to massage you, you can and should massage yourself. Sit on chairs with the knees lower than the hips and stay upright on your sit bones.

Manage your stress levels with walking and deep breathing. Focused breathing methods can greatly improve posture by aligning your spine from the inside out. The aim is to get rid of ingrained tension habits from slouching so that breath happens easily and naturally.

You can practice breathing through a straw while you sit on the edge of a bench or stool. Make sure you are on your sit bones with the curve in your lower back and abdominals relaxed. Keep the middle of your ear, shoulder, and hips aligned and practice keeping your shoulder blades drawing down as you breathe in through the straw.

After inhaling, hold your breath for a moment, take the straw out and slowly exhale making a SSSSS sound while focusing on keeping your waist area long and shoulders dropped during the exhale. Inhale again with the straw and notice how pulling the shoulder blades down will bring your breastbone up.

Again on the exhale, take the straw out and make a SSSS sound like a snake while you practice being on your sit bones and keeping length in your spine during the movements of exhalation. By practicing a lengthening of your trunk muscles when you exhale, you will teach your body how to enlist your abs to stabilize your spine rather than shorten and compress it with exercises that shorten your abs.

An easy way to improve your posture and your health

You can do this standing or sitting to get your body aligned from the inside out, turn on your intrinsic postural muscles and turn off all the extra work you have been doing by slouching.

Relax, walk around the room and notice how light your body feels and how easy it is to hold good posture and stop slouching by using the internal movements of breathing to activate your deep core.

Little time left to head off longshoremen's strike

NEW YORK (AP) ? Shipments of products as varied as flat-screen TVs, sneakers and snow shovels could sit idle at sea or get rerouted, at great time and expense, if more than 14,000 longshoremen go on strike as threatened ? a wide-ranging work stoppage that would immediately close cargo ports on the East Coast and the Gulf of Mexico to container ships.

Commerce could be brought to a near standstill at major ports from Boston to Houston if the strike takes place on Sunday, potentially delivering a big blow to retailers and manufacturers still struggling to find their footing in a weak economy.

"If the port shuts down, nothing moves in or out," said Jonathan Gold, vice president of supply chain and customs policy at the National Retail Federation. And when the workers do return, "it's going to take time to clear out that backlog, and we don't know how long that it's going to take."

The 15 ports involved in the labor dispute move more than 100 million tons of goods each year, or about 40 percent of the nation's containerized cargo traffic. Losing them to a shutdown, even for a few days, could cost the economy billions of dollars.

In addition to transporting goods, U.S. factories also rely on container ships for parts and raw materials, meaning supply lines for all sorts of products could be squeezed.

"The global economy moves by water, and shutting down container ports along the East and Gulf coasts while the national economy remains fragile benefits no one," Deborah Hadden, acting port director at Massport, the public agency that oversees shipping terminals in Boston. It is not a part of the contract dispute.

Florida Gov. Rick Scott said "the livelihood of thousands of Florida families lies in the balance."

The master contract between the International Longshoremen's Association and the U.S. Maritime Alliance, a group representing shipping lines, terminal operators and port associations, expired in September. The two sides agreed to extend it once already, for 90 days, but they have so far balked at extending it again when it expires at 12:01 a.m. Sunday.

The union said its members would agree to an extension only if the Maritime Alliance dropped a proposal to freeze the royalties workers get for every container they unload. The Alliance has argued that the longshoremen, who it said earn an average $124,138 per year in wages and benefits, are compensated well enough already.

Federal mediators have been trying to push negotiations along, but there has been no word from either side on the progress of the talks since Dec. 24. As recently as Dec. 19, the president of the longshoremen, Harold Daggett, said the talks weren't going well and that a strike was expected.

The work stoppage would not be absolute. Longshoremen would continue to handle military cargo, mail, passenger ships, non-containerized items like automobiles, and perishable commodities, like fresh food.

Joseph Ahlstrom, a professor at the State University of New York's Maritime College and a former cargo ship captain, called container ships the "lifeblood of the country."

"We don't fly in a lot of products. It's just too expensive," Ahlstrom said. "The bulk of the products we import come in inside containers."

The White House has weighed in on the issue, urging dockworkers and shipping companies Thursday to reach agreement "as quickly as possible" on a contract extension. Obama spokesman Matt Lehrich said the administration is monitoring the situation closely.

If it happens, the walkout could be the biggest national port disruption since 2002, when unionized dockworkers were locked out of 29 West Coast ports for 10 days because of a contract dispute.

The ports only reopened after President George W. Bush, invoking powers given to him by the 1947 Taft-Hartley Act, ordered an 80-day cooling-off period. Some economists estimated that each day of that lockout cost the U.S. economy $1 billion. It took months for the retail supply chain to fully recover.

An East Coast port freeze would have its biggest impact at the Port Authority of New York and New Jersey, where 3,250 longshoremen handled 32.3 million tons of cargo in 2010. The authority is not a party to the contract dispute.

Other major ports affected would include Savannah, Ga., which handled 18 million tons, and Houston and Hampton Roads, Va., which each handled more than 12.5 million tons.

Thousands of other jobs would be directly affected by the shutdown. Truck drivers might not have any cargo to transport, tug boat captains no ships to guide and freight train operators nothing to haul.

Simultaneously, another labor dispute involving dock workers was playing out on the West Coast.

Longshoremen at several Pacific Northwest grain terminals worked Thursday under contract terms they soundly rejected last weekend. The owners implemented the terms after declaring talks at an impasse. The International Longshore and Warehouse Union has yet to announce its next move.

Workplace rules, not salary and benefits, have been the obstacle to a new deal.

The dispute involves terminals in Portland, Ore., Vancouver, Wash., and Seattle, where longshoremen have been working without an agreement since the last contract expired Sept. 30.

Understanding cell organization to tackle cancer

Dec. 23, 2012 ? Scientists at The University of Manchester have identified how cells know which way up they need to be. The discovery could help in the fight against cancer because in the early stages of the disease the cells become disorganised.

Professor Charles Streuli and Dr Nasreen Akhtar of the Wellcome Trust Centre for Cell-Matrix Research have conducted new research that leads to a better understanding of cell polarity. Properly organised tissues are vital to maintaining functional organs and a healthy body. Part of being organised includes cells being in the correct position within the tissue and the right way up, because the top and bottom of cells have different functions.

The extracellular matrix (ECM) is a layer of protein rich material that surrounds tissues and helps to design and shape all of our organs. Previous studies have demonstrated that the ECM sticks to the cells and guides them into the right position. What hadn't been identified is how the ECM communicates that message.

To understand this better Professor Streuli and Dr Akhtar looked at epithelial cells, which make up the majority of tissues within the body. They studied epithelial cells of the breast, which make milk. These cells also form the linings of mammary ducts to carry milk towards the nipple. It's vital that these cells are organised correctly in order to make milk accessible for the baby. One of the first signs of cancer is that the epithelial cells become disorganised.

Breast epithelial cells connect to the ECM through receptors called integrins. In experiments using mice Professor Streuli and Dr Akhtar removed one of the genes responsible for integrins. They found that without that gene, the cells were both the wrong way round and in the wrong place so the breast tissue became disorganised. They then tried removing integrins in cultured cells from the breast, which produced the same effect of disorder.

Further study revealed that within the cell the integrin receptors connect to the protein ILK. This protein then links to microtubules, a network that forms the transport machinery of the cell. Integrins and microtubules ensure that inside the cell the correct proteins are transported to the top and the bottom of the cell.

The findings have been published in the journal Nature Cell Biology. Commenting on the research Professor Streuli says: "What we identified is a vital interplay between the transport machinery and the integrin receptors which makes sure that proteins are transported to the correct area of the cell. Without this interplay the proteins end up in the wrong place, and this can lead to cells becoming disorganised."

He continues: "What's really interesting is that when we compared breast tissue from our experiments with tissues of patients with early forms of breast cancer, they looked very similar. The cells were upside down and disorganised so they couldn't carry out their functions. We hope that our work to better understand cell polarity could ultimately lead to better diagnosis for cancer patients."

Whilst Professor Streuli and Dr Akhtar only looked at epithelial cells within the breast, they are confident their findings will translate to other organs. The accuracy of their experiments was greatly increased through the use of special 3D cultures to grow the cells, where they form tiny organs that look remarkably similar to real breast tissue.

Dr Akhtar explains: "Growing the breast cells so that they can form 3D structures rather than on hard petri dishes means they develop in a way that is much more akin to how they grow in the body. We were one of the first groups in the UK to be using this technology and we've been really pleased with the results."

Dr Akhtar has been working on this research for five years. She says: "I've been touched by cancer in my own family so I'm really passionate about understanding this devastating disease better. Over 90% of cancers come from epithelial cells, which is why we chose to study them. It's fundamental to understand how healthy cells work properly in order to fully appreciate why they go wrong when cancer develops, and how best to combat the disease."

The next stage of the research will be to investigate the link between altered levels of integrin and cancer, to determine whether this causes the disorganised nature of cells seen in the early stages of the disease.

Calif. woman charged in millionaire murder

A 22-year-old woman has been charged in connection with the high profile death of a 66-year-old millionaire who was tied up and then killed at his sprawling villa in Monte Sereno, Calif., last month.

The woman, Ravel Chanel Dixon, was arrested Tuesday for a separate crime ? offering sex, drugs or money to an undercover cop. On Friday, Dixon, of Alameda County, Calif., was charged with ?accessory after the fact? in the Nov. 30 killings of Raveesh Kumra, an investor who owned Western Cellular Management. Kumra is best known for owning Mountain Winery in the 1990s. ?

Police released a statement Friday saying they do not believe the homicide was a random act of violence.

Dixon?s charges carry a gang-enhancement, although prosecutors did not elaborate on what that meant. Her bail was set at $1 million at Friday?s court hearing.


Her mother, Sandra Mitchell, said outside court that her daughter, a single parent and college student, is scared. Mitchell called the charges "very troublesome."

John Ambrosio, Dixon's attorney, added:?"We don't know how they came up with these serious charges. It's an out-of-the-blue charge. They just want someone to charge that homicide to."

Dixon did not enter a plea and was told to return to court on Jan. 14.

Kumra's ex-wife, Harinder "Rani" Kumra, was also home and tied up during the attack. She has declined interview requests. The couple had divorced but remained cordial, according to a previous family statement.

Rani Kumra told police she believed perhaps three or four people had entered her family's massive home on Withey Road. She said she and her husband were bound at about 1:30 a.m.

Police have not explained how her husband was killed. Kumra was Mountain View?s first homicide victim in 40 years.

Also unclear is who killed Kumra. Dixon is not being charged with murdering him, and a sketch of a possible suspect in the case was released by Los Gatos-Monte Sereno police shortly after the homicide.

The sketch shows a skinny Latino or white man in his early 20s who stands about six feet and who has black hair.

The release added that department is "aware of the concern and anxiety this crime has caused the community and wishes to provide a limited update of the case."

Kumra's family issued previous statement through email, in which his brother, Bharat Kumra, described his brother as a man who "worked very hard and invested successfully over the years." He stated that his brother's wealth has been "overstated and inaccurate." Kumra had once owned the Mountain Winery in the 1990s and had invested in Tesla Capital.

"While he experienced success during his life, at the time of Ravi's passing he was weathering financial hardship," Bharat Kumra wrote. "He was a generous man and went out of his way to help anyone he met, even when he had little."

Kumra had a minor criminal past in Santa Clara County - he was charged with several misdemeanors and one felony in the 1990s, court records indicate. He was arrested in connection with a felony drunk-driving incident in 1995. In 1998, he was charged with assaulting a security guard at the Mountain Winery and making a threatening phone call to the then-executive director of the Villa Montalvo, an arts foundation.

Kumra was born in Kartharpur, India and immigrated to the U.S. in 1970 after earning a degree in chemical engineering from Indian Institute of Technology Delhi, according to his obituary. He eventually settled in the Bay Area. His family said he was one of the first Indian-born entrepreneurs in the wireless industry.

WHO ate all the pies? In 6th-century Jerusalem, the Byzantine monks were greedy gobblers - despite strict rules that they should eat mainly bread and water.

Most early Byzantine monasteries were located in remote deserts, but St Stephen's monastery thrived in Jerusalem. Wondering how urban living affected the monks, Lesley Gregoricka at the University of South Alabama in Mobile took bone samples from 55 skeletons buried under the monastery.

The ratios of various isotopes of carbon and nitrogen in the bones confirmed that the monks ate a lot of common cereals like wheat, as well as fruit and vegetables. But many bones were rich in the heavy isotope nitrogen-15, suggesting the monks ate lots of animal protein. That could mean meat, or dairy products such as cheese (Journal of Anthropological Archaeology, doi.org/jzt).

"The rules on issues such as poverty, chastity and obedience were certainly known and could not be easily ignored," says Peter Hatlie of the University of Dallas's Rome Program in Frattocchie, Italy. "Only fallen, weak, mad and demonic monks ate meat."

New pathways that drive metastatic prostate cancer identified

Dec. 21, 2012 ? Elevated levels of Cyclin D1b could function as a novel biomarker of lethal metastatic disease in prostate cancer patients, according to a pre-clinical study published ahead of print on December 21 in the Journal of Clinical Investigation by researchers at the Kimmel Cancer Center at Jefferson.

The group, headed by Karen E. Knudsen, Ph.D., Professor and Hilary Koprowski Chair, Departments of Cancer Biology, Urology, and Radiation Oncology at Thomas Jefferson University and Deputy Director for Basic Science at the KCC, found that Cyclin D1b, a variant of the cell cycle regulator Cyclin D1a, functions independently of the cell cycle to promote metastasis in both early and late stage prostate cancer.

Rather, Cyclin D1b, but not Cyclin D1a, regulates a large gene network, the researchers found, which was shown to cooperate with androgen receptor (AR) signaling to fuel metastatic progression in multiple models of prostate cancer.

Studies have shown that Cyclin D1b expression is elevated in early stages of prostate cancer (in up to 30% of primary disease), and researchers have now demonstrated that this occurs more frequently in late stage castration-resistant prostate cancer: up to 80%.

Cyclin D1b expression is also highly correlated with that of the pro-metastatic gene SNAI2 (Slug), which the group identified as regulated by cooperative signaling between Cyclin D1b and AR.

"Numerous clinical and pre-clinical studies have effectively demonstrated that AR signaling is critical for progression to metastatic disease, but our knowledge of AR targets which can induce metastatic phenotypes is limited," said Dr. Knudsen. "Our data describe how cross talk between the cell cycle and AR can rewire the AR signaling axis to enhance the expression of genes which elicit metastasis in both early and castration resistant prostate cancer models."

"We found that Cyclin D1b can directly promote AR dependent expression of the gene SNAI2 (Slug), which dramatically increased metastatic events to soft tissues in animal models," she added.

Metastatic castration resistant prostate cancer represents the most lethal form of the disease, which arises when AR is reactivated despite continued hormone therapy.

Soft tissue metastasis to the liver and lung represents a particularly aggressive form of prostate cancer, whose presence predicts for decreased survival time in prostate cancer patients.

Currently, there is little knowledge as to how these metastatic events occur, and identification of pathways and biomarkers of this lethal event could greatly benefit prostate cancer patients.

Using various in vitro and in vivo models, researchers found that Slug enhances the ability of cells to colonize soft tissues, which resulted in a higher incidence of metastasis in the liver and lung.

Given the inability to manage AR signaling in metastatic castration resistant prostate cancer, Slug driven pathways could be leveraged to dramatically limit the incidence of soft tissue metastasis and improve patient morbidity and mortality, researchers believe.

"Identification of AR driven pathways which mediate metastatic progression represents a significant leap forward in our attempts to effectively manage prostate cancer progression," said Dr. Knudsen. "Cyclin D1b and Slug can likely be used as biomarkers to identify patients with an increased risk of metastasis, and will eventually provide us with novel "druggable" targets downstream of AR and Slug which can be exploited to dramatically reduce the incidence of these lethal metastatic tumors."

UK man jailed over forged Churchill signatures

LONDON (AP) ? A British antiques dealer has been jailed for 10 months for forging the signatures of Winston Churchill and other famous figures in books he sold as collector items.

Prosecutors say Allan Formhals bought books at recycling centers and junk sales and made thousands of pounds selling them on eBay after adding the bogus autographs.

Judge Peter Henry said police who searched the 66-year-old's home in Milford-on-Sea in southern England found "an Aladdin's Cave" of forged signatures from Oliver Cromwell, Queen Elizabeth I, Marie Antoinette and others.

Formhals was convicted in October of 10 counts of fraud and sentenced Friday.

Ian Lawson of the Metropolitan Police economic crimes unit said police had recovered more than 100 forged volumes, but many more are believed still to be on the market.

Former IMF boss 'pimping' case to go ahead

The prosecution of former International Monetary Fund chief Dominique Strauss-Kahn for pimping is to go ahead, French judges ruled Wednesday.

The ruling was given just over a week after Strauss-Kahn settled a separate civil case in New York with a hotel maid who accused him of attempted rape in May 2011, ending his French presidential hopes and career at the IMF.

While the New York settlement brought his U.S. legal woes to an end, the latest decision by the court in Douai in northern France means he remains under the legal spotlight at home.

"Dominique Strauss-Kahn's defense team is certain that he will ultimately be cleared of these absurd accusations of pimping," lawyer Henri Leclerc said in a statement Wednesday, adding that he planned to appeal to France's supreme court.

Strauss-Kahn denies wrongdoing in all the charges against him.

He is under fire about sex parties with prostitutes in the so-called Carlton Affair, named after a hotel in northern France at the center of the inquiry.

His lawyers argue that consorting with prostitutes is not illegal and that investigators have no grounds for pursuing him on the basis that his behavior could be construed as pimping, which is illegal.

Under French law, prosecution does not automatically lead to trial, but it often takes months or years before a decision is made.

In the United States, Strauss-Kahn's legal troubles ended within 18 months of a sexual-assault complaint filed by New York Sofitel hotel maid Nafissatou Diallo.

U.S. prosecutors dropped criminal charges in August 2011, saying they had worries about Guinea-born Diallo's credibility as a witness in court after discovering that she had lied in the past on tax and immigration documents. She opened civil proceedings that ended last week with a settlement for an undisclosed sum.

A former New York hotel maid has settled her sexual assault lawsuit against former International Monetary Fund Chief Dominique Strauss-Kahn, although the amount of the settlement was kept confidential. TODAY's Natalie Morales reports.
